Pouille displayed vast nerve in staging a 6-1, 2-6, 6-4, 3-6, 7-6 (6) win against Nadal in a four-hour marathon that was decided in the tie-breaker of the fifth set.
He won the next two points to earn a thrilling 6-1, 2-6, 6-4, 3-6, 7-6 (8-6) fourth-round upset of Nadal in 4hr 8min.
He led by a break at 1-0 and 4-2 in the fifth set, but Pouille broke back to 4-all, and eventually converted his fourth match point in the tiebreaker with a gutsy forehand victor to close a 16-stroke exchange.
The 22-year-old then pulled 6-3 clear to open up three match points but, while Nadal saved them all, he then missed a simple forehand and Pouille pounced to snatch a momentous victory. "I was very aggressive all the match and at 6-6 [in the tiebreak] I didn't want to miss", he said after the match.
However, with the court at his mercy, Nadal fired the forehand into the net, giving Pouille a 7-6 lead, which the 24 seed would not relinquish, hitting a victor down the line to seal victory.
Top seed Novak Djokovic, starved for competitive action, feasted against unseeded Briton Kyle Edmund, gobbling up a large serving of points in a 6-2 6-1 6-4 romp on Sunday that put him into the U.S. Open quarter-finals.
Nadal and Pouille had only played once before, and Nadal won.
"Was a big mistake", he said.
The 24th-seeded Pouille joined No. 9 Jo-Wilfried Tsonga and No. 10 Gael Monfils with fourth-round victories Sunday.
"Is true that I don't have lot of matches on my shoulders for the last three, four months, but even like this I lost an opportunity", Nadal said.
Novak Djokovic, of Serbia, hits a return shot to Kyle Edmund, of Britain, during the fourth round of the U.S. Open tennis tournament, Sunday, Sept. 4, 2016, in NY.
Anastasija Sevastova, Latvia, def. Johanna Konta (13), Britain, 6-4, 7-5. "I [fought] right up to the last ball", said Nadal. By reaching the last eight Kerber has put herself in position to end Serena Williams' long reign as world number one. That's the truth. I lost an opportunity to have a very good event here. And after winning at least one Grand Slam title each year for a decade, he's now gone two full seasons without one. Beating Nadal, a 14-time Grand Slam champion, is no easy feat. He then got a chance to rest the arm, because his second-round opponent withdrew before the match, and his third-round opponent stopped after only six games.
The defending champion will now face Tsonga in the quarterfinals, while Monfils plays Nadal's conqueror Pouille. The young Frenchman held his nerves in the tiebreaker and advanced to the quarterfinals with a 6-1, 2-6,6-4, 3-6, 7-6 (6) win. "Was tough. And now I need to come back there", he added.
Sevastova, who retired in 2013 but made a comeback, will take on Caroline Wozniacki of Denmark after she overcame American Madison Keys, seeded eighth, 6-3, 6-4.
Nadal has enjoyed greater success away from the grand slams this year but he rejected the suggestion he is struggling to cope with the pressure.
